Thi s inst ruct ion manual has been prepared f or the Oly mpus Invert ed Research Microscope Model
IMT. I t is recommended that you read the manual caref ull y in order to famil iar ize yourself f ull y
wi t h your microscope in order to obt ai n opt i mum performance f rom t hi s precision inst rument .
IMPORTANT
Observe the following points car efully:
Operation
1. A lways handle t he microscope wit h the care i t deserves, and avoid abrupt motions.
2. Avoid exposure of the microscope t o direct sunlight, dust and vi brati on.
3. Only use the tensi on adj ust ment ring for altering the t ension of th e coarse adj ust ment knobs.
Do not twist the two coarse adjustment knobs in th e op posite directi on s si mult aneously,
as th is causes damage.
4. Ascertain t hat t he voltage select or swi tc h on th e base plate is set t o conform wi t h t he l ocal
mains voltage.
5. Di sconn ect the li ne cord fr om the AC power out let for fu se repl acement .
6. Object ives provided to t he Mod el IMT are designed for observat ion of speci mens t hrough
cult ure vesse ls of 1mm th ickness. Therefore, be sure to use cult ure vessels, th e bottom of
wh ich is 1 mm O.3 t hick and evenly f lat .
Maintenance
1. Lenses must always be kept clean. Fine dust o n lesn surfaces sho uld be blown or wiped off
by means of a hand bl ower or a clean brush. Carefully wipe off oil o r fingerp rin t s deposited
on t he lens surf aces wi t h gauze moi stened with a small amount of xy l ene, al coh ol -et her
mixture (7 : 3) or et her.
2. Do not use organic sol utions t o wi pe t he surfaces of vari ous compon ents. Plast ic part s,
especi all y, should be cleaned wi t h a neut ral detergent.
3. Never di sassemble the mi croscope f or repair .
4. The microscope shoul d be st ored in it s contai ner immed iat ely aft er use. If this is not
possible, it shou ld be covered with the vi ny l dust cover provided . It is best to keep objectives
and eyepieces in desiccat ors. contai ni ng desiccants such as si li ca gel.

Rotate t he ring t o stabilize t he base.
NOTE 1: The lower end of t he pill ar has two notches (l arger one at t he fr ont and smaller one
at t he back ) and a connect i ng cord comes out of it . Insert th e connect i ng cord and
l ower end of t he pi ll ar into th e base hol e vertically and clamp th e pill ar with screw at
t he bottom of t he base. The pillar direct ion is determi ned by f i tting t he notches so
that t he connect i ng cord passes through t he larger not ch and connects t o the
receptacle on the left hand side of the base back .

NOTE 2: Th e mechanical stage is attachable t o the right or left hand side of t he plai n st age and
clamped with a coi n. I f t he mechanical stage is attached, only one extension pl at e can
be attached to t he opposite side of t he plain stage.
@ How t o mount t he mechanical stage:
Fig. 1 shows t he mechanical stage attached on t he ri ght hand side of t he plai n stage. I t
is possibl e to attach the mechanic al stage on t he left side as shown i n Fig. 2 ac cord ing
t o your prefere nce. To correct ly attach t he mechan ical staqeon eit her side, al ign t he
edge of t he mechanical st age wi t h the plai n stage corner as indicated by arrow s.

It is necessary t o mount t he opti onal aux ili ary lens IMTA L t o t he lamp house
support and attach the condenser and con denser holder t o the stage. The lower
edge of t he suppor t should be aligned wi t h t he red l ine on t he pi l lar.
* For observat ion wi t h object ives 40x or higher , use an ordi nary obj ect ive and put
a cover glass under t he speci men.
d) For use of the contrast attachment BHNIC:
It is necessary to ali gn t he lower edge of t he support wit h the bl ack index li ne on
the pillar, and use t he auxi li ary lens IMT- AL.
* Only a cover gl ass of 0.17 mm t hick can be used.

SUMMARY OF PUTTING THE MICROSCOPE IN OPERATION

purpose (page 5).
E. Place a specime n on th e stage (page 10 ).
F. Coarse focus with th e lOX object ive.
* If necessary, stop down the aperture diaphra gm to r easier focusing.

G. Make interpupi llary and di opter adju stments (page 11 ).

V. OPERATION
A. Electric Equi pment
1. Adju stmen t of Minimum V ol t age
The minimum voltage req u ir ed fo r t he ligh t sour ce can be adj ust ed w i t h t he rh eost at
tr immer screw at th e mi croscope base plat e in accordance with t he li ne voltage and
f requency . Th e bu il t- i n rheostat incorporates a thyr ist or i n i ts semiconductor c i rcu it for
the following advantages :
a) Ext remel y fi ne adj ustment of l igh t intensity.
b) El imination of f l ick er and stabi l izat ion of l igh t
int ensi t y .
c) Increased l if e expect ancy of the bul b.
For adjustment of t he minimum li ne vo lt age, ascer-
tain that the voltage selector swi tch is set to con form
with t he local mains volt aqe, and t he slidi ng co ntrol
lever is positi oned closest to you (l ow vo! taqcl ,
and then activate the mai n switch CD . I f th e bu lb is Fig. 5
di mly lit, t he seconda ry vol t age is cor rect. I f it is
not l it at al l, rot at e t he rheost at t r i mmer screw gradual ly wit h a co i n, unti l t he bulb is

2. 30W Tun gst en Bulb
The standard li ght source i ncorporates a 30W pre-centered tu ngst en fil ament bul b,
pr ovided wi t h a socket for pos it ive cont act , el iminati ng t he problems of defect ive contact
and over -heat i ng. When used at t he sett ing " 6" on t he control lever scale, th e average
l ife of t he bu lb LS30 is lon ger t han 200 hour s. Thi s is, however, greatly reduced, if t he
bulb is used at higher voltage. Theref or e, it is advi sable t o avoid prolonged use at readi ngs
over "6" (in t he red zone).
* It is recommended t o avoid swi t ching on and off the t ungsten bulb wit h t he sl idi ng
control lever at high i nt ensit y position (away fr om the user ). It reduces bulb life.
B. Specimen Hol ders

After coarse and fine adj ust ments, rotate t he correct ion col lar . keeping t he specimen in
fine focus until opt i mum resoluti on is obtai ned. A proper use of th is coll ar is specia lly
ef fecti ve t o prevent the deteri ora tion o f t he objective resol uti on to be caused by t he un -
even th icknesses of various pet ri dishes, cult ure bottl es, etc .
C. Observat i on Tubes
1. Interpu pill ary Dist ance and Diop t er Adjustments
1) Hol d the knurl ed sli di ng grips <D of th e ri ght
and left eyepiece tubes wit h both hands and push
t he t ubes toget her or pull th em apart lat erall y ,
wh ichever is required, while looking t hroug h th e
eyepi eces wit h bot h eyes, until perfect binocular
vi son is ob tai ned. (F ig. 6)
2) Rotate the tube length adju stment ring on t he
right eyepiece tu be t o match your interp up i l lary
distance setti ng whci h yo u obtai ned from th e
scale .

4) Next, lookin g at t he image through the left eyepi ece with your left eye , rotate t he
tube length adjustment ring @ t o f ocus on the speci men wi t hout using th e coarse
and fi ne adj ust ment knobs.
2. Eyepi ece Cap
The eyep iece cap is recommended f or t hose who wear eyeglasses. It prevent s scrat ching
t he eyeglasses.
D. Ill uminat ion f or Phase Cont rast Microscopy
1. Ult ra Long Work i ng Di st ance Il l uminat ion
The t otal work i ng di stan ce is 55mm. long enough
to permit observation of large size culture bott les.
1) Move the phase annu lus sli de CD unt i l the rnaq-
nif icat ion i ndex engraved on th e slide matches
th e ob ject ive magni fica tion i n use (F ig. 7) .
* Make sure th at the sli de cl ick s int o position .
2) Place a specimen on th e stage and bring it into
focus.
NOTE: It is suggest ed t o stop down the field i ri s
idaphragm for easier fo cusing . ( Fig. 8)
After f ocusing, op en th e full aper ture @ .
3) Insert the li ght annul us into th e auxiliary lens.
NOTE: Use t he l ight annul us t hat mat ches the
object ive magnifi cat ion.

4) Remo ve one of the eyepi eces from t he observat ion t ubes and insert the cent er ing
telescope int o t he observat ion tube .
5) Rotate t he top lens assembly of t he center ing telescope to bri ng the br ight r i ng ( l ight
annul us) and the dar k r ing (phase annulus) int o focus.
6) Operate the center i ng screws of the li ght annulus unti l both annul i are concentric
and superi mposed. (Fig. 9)

7) Remove t he cent er i ng t elescope and insert t he eyepiece bac k into t he observat ion
tube. Now you can st art you r phase cont rast observatio n.
NOTE: 1. Repeat the procedure t hrough 1) to 6) above each t ime objectives are
changed.
2. In case of ul t ra long worki ng dist ance ill uminat ion, t he fi eld i r is diaphragm
@ in Fig. 8 is used as apert ure diaphragm.
2. Long Wor king Dist ance Illum inat ion
The total wo r ki ng dist ance is 20 mm so long as t o per mit observation of large size Petri
dishes.
1) Set th e tu rret condenser I MT-LWCD at the " 0" posi ti on.
2) Coarse f ocus with the 10 X object ive.
NOTE: It is suggest ed t o stop down sli ghtly t he apert ure i ri s dia phragm @ fo r easier
f ocusing (F ig. 10)
3) St op down the f ield i r is diaphragm t o the mini mum by means of the lever CD . Rotat e
t he condenser height adjustment kn ob unt i l t he i mage of t he f ield d iaphragm
is visibl e sharply in the field of view.

4) Br ing the image of the f ield diaphragm int o the center of the f iel d by means of the
condenser centeri ng screw (Fi g. 10) . Re-open the d iaph ragm until the small r ing
of the di aphragm becomes a larger pol ygonal r ing around the ci rcular edge of the f ield
(Fi g. 11).

5) For phase cont rast observat ion, open t he apert ure di aphragm fu ll y .
6) Rotate the turret condenser until the light annulus " ' 0" is in posit ion.
7) Match the phase annulus sl ide at the " 10" positi on (F ig. 7).
8) Insert the centering telescope CT into the observation tube after removal of the eve-
piece. and rotate the top lens assembly of the CT to bring the l ight annulus and phase
annulus into focus.
9) Operat e the center i ng screws of the li ght annulus (Fig. 10) until l ight annulus and
phase annulus are concentric and superimposed.
* After complete cent rat ion, clamp the centering screws with care.
10) Remove the CT and insert the eyepiece back into the observation tube. Now it is
ready for phase contrast observation .
NOTE : Mat ch the light annulus and phase annul us to the objecti ve magnif icat ion
whenever object ives are changed, but re-centr ati on is not necessary.
E. Field and Apert ure Iris Diaphragms
1. Field Iris Diaphr agm
The field iris diaphr agm controls the diameter of the ray bundle impinging on the speci-
men surface and thus increases image defin ition and reduces glare.
2. Aperture Iri s Diaphragm
In order to achieve optimum obj ecti ve perf ormance in brightfield. the opening of the
aperture iris diaphragm should be mateched to t he numerical aperture of the object ive in
use. I t is often preferable, however, t o stop down the aperture diaphragm by about 70%
to BO% of the object ive N.A. (Fig. 121.
During phase contrast microscopy. however. the aperture iris diaphragm should be kept at
fu ll ope ni ng.

F. Focusing Adjustment
1. Tension Adjustment of Coarse Adjustment Knobs
A t ension adjust ment ring CD is provided next t o t he
right hand coarse adjust ment knob. With thi s device
t he tension of t he coarse adj ustment is freely adj usta-
ble for either heavy or l ight movemen t , dependi ng on
operator preference (F ig. 13l. However , do not
loosen t he tension adjust ment r ing too mu ch, because
the stage drops or t he f i ne adjustment knobs sl ip
easily . I f i t recommended to tig hten t he r i ng especial -
ly f or prolonged observat io n or cincphot ornlcro-
qrap hv .
NOTE: Be caref ul not t o rota te t he right and lef t
coarse adjust ment knobs i n t he opposite
di rect ions si mul taneously.
2. Automati c Pre-focusing Lever
Th is lever CD is provided to prevent possible cont act
bet ween specimen and obj ecti ve as well as to simpl if y
coar se f ocusing. The lever is locked after coarse focus
has been accomplished. This prevents f urt her down-
ward t ravel of the stage by means of t he coarse
adjust ment knobs and automat ical ly provides a li mit -
ing stop if the stage is raised and t hen lowered again.

3. Adjustment of St age Block Height
The stage bl ock shou ld be al igned wi t h th e stage bi ock height index " B" , engraved on
the microscope stand (Fig. 14) for bi ological and tissue culture observat ion.
Beside the vert ical movement of t he stage by means of coarse and fine adj ust ments, th e
stage block hei ght can be changed upwa rd af t er loosening two stage block loc k i ng levers

careful not to hit the objective front lens against the stage by rotating the
nosepiece. Make it a point to hold the st age block steady with your hands
while you change the stage block height when the lower limit st op pin is re-
mo ved .
