Divisions, So Each Division Has 0.001mm.: Parasitology Notes (Laboratory)

This document discusses different tools used to measure microscopic organisms: 1) A stage micrometer is etched onto a glass slide with known divisions of 0.001mm and is used to calibrate the size scale of a microscope. 2) An eyepiece graticule or ocular micrometer has a scale fitted in the eyepiece that does not change with magnification and can be used to directly measure microscopic objects. 3) A hemocytometer has a grid of 9 squares, each 1mm by 1mm, divided into spaces; it is used with formulas to calculate the size of objects viewed through a microscope.

Parasitology Notes (Laboratory) NOTE: Stage micrometer has 100

divisions, so each division has

0.001mm.
 Micrometry
-measurement of microorganism
-determination of size of objects by aid of a scale
calibrated with a stage micrometer.
-Measures the length, width and thickness of the
microorganism.
 Meter: 1mm = 0.001m
 Micrometer: 1µm = 0.001 mm = 0.000001m  Hemocytometer
 Nanometer: 1nm = 0.001µm = 0.000000001m -each scale is 3mm wide and 3mm long
 Eyepiece graticule or Ocular -whole scale is divided into 9 big squares
Micrometer
-each square is 1mm long and 1 mm wide
-scale fits in eyepiece of a microscope that can be
-1 hemocytometer space = 0.05
used to measure microscopic objects.
-do not change even when image was magnify.
-numbered
-the higher the length of ocular spaces, the more
accurate.

Formulas:

= value of one ocular

No. of stage spaces x 0.01 x 1000 space
No. of ocular spaces in micra

 Stage Micrometer
-small ruler etched onto a glass slide that has
known lenght of 1mm. = value of one
No. of hemacytometer spaces x 0.05 x 1000 ocular space
No. of ocular spaces in micra
