SOFTWARE PASSWORD
This password allows you to enter the security section of the software used on your spectrophotometer.
Through the security section, you can “lock” test setups (test parameters) so that they may not be altered.
The password also allows you to remove the security so that you may edit the test parameters. Please
refer to the appropriate section in your Operator’s manual for more information on locking a test.

Chapter 3 Troubleshooting Guide
The Troubleshooting guide is intended to give the experienced spectrophotometer repairperson a starting
point to repair a spectrophotometer. It is not intended to be a guide for the average user. The Initialization
Sequence will help isolate problems encountered during instrument startup. The Troubleshooting guide
contains both previously seen problems, and contrived cause-effect problems; e.g., what happens when a
motor is unplugged. The important thing to remember with this instrument is that the sample detector is
used to initialize the entire instrument. This means that anything that keeps light from getting to the
detector, such as a problem filter wheel, or something in the sample compartment will keep the instrument
from initializing properly. The detector or Main board could cause a similar problem if there is something
wrong with the electronics.
Initialization Sequence
1. Power on.
2. Calibrate turret
a. Rotate the turret while reading the turret sensor.
b. After we have detected 3 small openings without finding the big opening, reverse the
turret rotation direction.
c. After we have detected 6 more small openings without finding the big opening, return a
turret calibration failure error.
d. When we find the big opening (indicates position B), center the turret in the opening.

8. On the VIS instrument, move to 681nm and determine the ADC integration time.
9. Move to the last wavelength used on the walkup (ATC) screen, and zero the system.

Operating the instrument with the cover off exposes the operator to potentially dangerous
voltages and ultraviolet (UV) radiation. Therefore, it is recommended that only authorized
service representatives perform procedures requiring removal of the instrument cover and
replacement of electrical components. To protect both yourself and the instrument, be
sure to contact an authorized service representative to perform any service procedure not
listed in this section.
Routine care
Routine care for your spectrophotometer does not require a lot of time. To help minimize maintenance
time and to increase the life and performance of your instrument, please follow these guidelines:
• To prevent dust from accumulating on and in the instrument, always replace the dust cover when the
instrument is not turned on. The dust cover, supplied with the instrument, is resistant to most aqueous
solutions.
• Gently wipe the outside of the instrument with a soft cloth to remove any dust or spills. Water,
isopropyl alcohol and other common laboratory-cleaning agents may be used if necessary.
• Always clean up spills to prevent or minimize damage to the instrument. If concentrated acids or
bases, or any hydrocarbon materials, are spilled on the instrument, be sure to clean up the affected
area immediately.
• Use water, alcohol or other common laboratory-cleaning agents to clean the keyboard. It is
recommended that you clean spills off the keyboard as soon as they occur.
Cleaning
Cleaning and maintenance of cells
Cleaning of cells both inside and out is important not only because any contaminating material may
absorb light, but also because material within the cell may react chemically with subsequent reagents or
standards introduced into the cell. Cleaning methods depend to some extent on the nature of the
contaminating material. Sodium (or ammonium) hydroxide and dilute hydrochloric acid may be used to
remove some acidic and basic contaminants, respectively. Clorox® (undiluted or 1:1) is very effective in
removing proteinaceous and bacterial contaminants. You may also use Cell Cleaning Solution
(332260-169) to clean your cells.

Handle and dispose of chromic acid with care!
Finally, soaking in chromic acid will remove most contaminants, but the acid should be handled, and
disposed of, with care. Because of the exothermic reaction of the acid and water, any heat generated
should be quickly dissipated to avoid altering the pathlength of the cell. Cells should not be placed in hot
chromic acid.
Note: To prepare chromic acid cleaning solution, slowly add (with stirring) 800mL of
concentrated sulfuric acid to 458mL of distilled water containing 92g of sodium
dichromate (Na2Cr2O7•2H2O). This cleaning solution should be red-brown. Discard, using
proper disposal methods, when green tinge appears.
Cells with scratches in the optical path should be discarded because scratches will cause anomalous
absorbance readings. Cells should be protected during cleaning and never thrown into a bath with
glassware where they might get scratched, or placed in a wire rack that might scratch them. The outside
of the cells may be wiped with a soft, lint-free tissue, and should be kept free of fingerprints.
Micro Flowcells
• Aspirating dilute acid, base, non-filming detergent or Clorox through the cell in short bursts
• Do not use acetone to clean the windows of the sample compartment. Instead, use a non-abrasive
laboratory cleaning solution (Cell Cleaning Solution, 332260-169), distilled water or alcohol.
• Use the liquid and a soft, lint-free cloth to clean the windows. Do not apply too much pressure or
the surface of the windows may be damaged. Be sure to remove all fingerprints.

WARNING
The lamp gets very hot during operation. Before removing the lamp, turn off the instrument
and allow the lamp to cool down for 10 minutes.

Turn off and unplug the instrument before removing the cover of the lamp compartment.

To avoid personal injury or damage to the instrument, follow these instructions carefully!
2. Make sure that the lamp has had time (at least 10 minutes) to cool off.

3. Loosen the captive screw (#1, Figure 4.1) on the lamp door by rotating it counterclockwise about ¼
turn.
5. Unscrew the thumbscrew (#2, Figure 4.1) holding the lamp assembly (#3, Figure 4.1) in place.
6. Carefully pull the assembly out until you can reach the connector.
7. Release the tab (#4, Figure 4.1) and disconnect the cable.
WARNING
Do not touch the lamp with your fingers!
9. Use a soft, lint-free cloth to wipe off any fingerprints on the lamp.
11. Position the new assembly as shown in #5 of Figure 4.1, making sure to align the pins correctly.
14. Plug the instrument into an appropriate wall outlet and turn on the power.
15. Press the UTILITY key. The Utility screen appears on the display.
16. Press the arrow keys to highlight Lamp Hours and press ENTER.

The fuse is located in the power entry module located at the center of the back panel of the instrument
(#1, Figure 4.2).
WARNING
The instrument fuse must be replaced with the same type and rating fuse.

If the fuse fails repeatedly, it may indicate a serious problem with the instrument. Contact
technical support as soon as possible.

2. Position the instrument so you can access the power entry module on the back of the instrument.
3. Remove the power cord. Insert a flat-blade screwdriver into the notch (Figure 4.3) on the fuse
cover and pry off the cover.

When servicing your spectrophotometer, always observe proper safety precautions.
USE CAUTION WHEN WORKING ON OR NEAR THE LAMP POWER SUPPLY. HIGH VOLTAGE IS
PRESENT.
DO NOT USE STEEL WOOL, EMERY CLOTH, SANDPAPER, OR FILES WHEN CLEANING THE
INSTRUMENT.
External Covers
WARNING
When in operation, certain internal components, such as the lamp and Power Supply,
become hot and may still be hot when the unit is opened for servicing. Avoid touching these
areas. Failure to observe this warning may result in burns.
NOTE: Before removing the covers, make sure all samples and sample holders have been
removed from the spectrophotometer.
• Open the cover over the sample compartment and remove any sample cuvettes.

Top cover
Removal
1. Access the back of the instrument and loosen the two screws at the back of the instrument (refer to
Figure 5.3). It is not necessary to remove the screws.
2. Lift up on the front of the instrument and press the two tabs outward to release them from their clips
(refer to Figure 5.4). While holding the tabs outward, press the tabs up through the bottom of the
instrument.
CAUTION: THE DISPLAY AND KEYPAD CABLES ARE CONNECTED BETWEEN THE
COVER AND THE MAIN CIRCUIT BOARD IN THE INSTRUMENT. BE
CAREFUL WHEN LIFTING OFF THE COVER TO MAKE SURE THE CABLES

3. Carefully remove the instrument cover and turn it upright ¼ turn so the cover can be set on its back
surface.
NOTE: The following steps are necessary only when replacing the display or keypad, when
replacing the Main Circuit board, or when it is desired to have freer access to the unit by
moving the top cover away from the instrument.
4. Open the ferrite bead, or the wire clip and release the cables.
5. Disconnect the connector for the display from connector J9 of the Main Circuit board.
6. Disconnect the connector for the keypad from connector J13 of the Main Circuit board.
7. Disconnect the printer cable from connector J11 of the Main Circuit board.
Replacement
1. If they were disconnected, connect the printer cable to J11, the keypad cable to J13, and the
display cables to J9 on the Main Circuit board. (All connectors are unique, so each cable can only
correctly attach to one connector on the Main Circuit board.)
2. Carefully insert the cables into the ferrite bead, or cable holder and snap closed.
3. Carefully position the top cover over the instrument by rotating it over the instrument. Make sure
the mounting screws are aligned with the slots for the screws and the tabs are aligned with the
holes in the bottom of the instrument.

NOTE: Make sure that you do not strain the keypad cable when moving the cover. This could
cause the cable to tear at the keypad.
NOTE: Make sure that the cover is not pinching any wires when it is setting on the base.
4. With the cover’s tabs aligned over the tab slots in the base, press down on the front right corner of
the cover until the you feel the tab protruding through the tab slot in the bottom of the instrument.
5. Push the cover to the left until you feel the front left corner tab protrude through the tab slot in the
bottom of the instrument.
6. Press the cover straight down until the tabs lock into place.
7. Access the rear of the instrument and tighten the two mounting screws (refer to Figure 5.3).
Internal Assemblies
NOTE: Prior to working on any of the internal assemblies, it is necessary to remove either the
bottom cover (refer to Bottom cover [Main board], Bottom cover [Power Supply], and Top
Cover).

2. From the topside, remove the two screws, which secure the limit switch to the base.
NOTE: Ensure that you do not touch the grating with the tool or with your hand.
3. Remove the solder where the wire assembly is connected to the terminals of the limit switch.
Replacement
2. Position the limit switch with its actuator button positioned as shown in Figure 5.4. Make sure the
grating arm makes proper contact with the limit switch at the end of the arm's rotation.
3. Insert and tighten the two screws, which secure the limit switch.
4. Route the limit switch wiring through the channel in the base so that the Monochromator cover will
not pinch the wires.
5. When all other work is complete, replace the Monochromator cover (refer to “Monochromator
cover").
NOTE: Replacing the limit switch does require new instrument calibration.

3. Lift the filter wheel out of its location, noting the orientation of the filter wheel, and remove the
screw and washers. Do not change the orientation of the spring washer.
Replacement
1. Insert the screw with washers through the center hole of the filter wheel making sure the orientation
of the wheel is proper so it will mesh with the drive gear.
3. When all other work is complete, replace the Monochromator cover (refer to "Monochromator ").
Removal
1. Remove the Beam Splitter and turret cover (refer to "Beam splitter and turret cover ").
NOTE: One of the screws for the Sample Detector cover is common to the Beam Splitter and
turret cover and is removed when that cover is removed.

3. Remove the remaining two screws securing the Sample Detector cover.
4. Lift the Sample Detector cover off the base and set aside.
5. Remove the Sample Detector Circuit board (335901-6034 [UV] or 335901-6044 [Vis]).
6. Remove the two screws securing the Sample Detector Circuit board to the base.
7. Carefully pull the Sample Detector Circuit board away from its locating dowel pins and remove it
from its mounting position.
8. Remove the ribbon cable from its connector on the Sample Detector Circuit board.
9. Remove the lens (335901-020S [Vis] or 335901-021S [UV]) if cleaning, or replacement is required.
10. Hold the lens with a lens tissue, soft cloth, or lab gloves and remove the two screws and washers
securing the lens.
CAUTION: NEVER TOUCH THE LENS WITH YOUR FINGERS. ALWAYS USE A LENS
TISSUE, OR SOFT COTTON CLOTH, OR CLEAN LAB GLOVES TO AVOID
OILS GETTING ONTO THE LENS.

NOTE: The lens does not have an orientation. It fits into a slight ridge, which positions it, and is
clamped in place by two washers, which are secured by screws on either side of the lens.
Replacement
CAUTION: NEVER TOUCH THE LENS WITH YOUR FINGERS. ALWAYS USE AN
UNCOATED LENS TISSUE OR SOFT COTTON CLOTH TO AVOID OILS
GETTING ONTO THE LENS. NEVER USE LENS PAPER WITH SILICONE
ADDITIVES.
2. Hold the lens with a lens tissue, soft cloth, or lab gloves and position the lens on the ridge in the
base.
3. Insert the screws through the washers and gently snug them up against the lens to hold it in place.
DO NOT OVERTIGHTEN OR THE LENS MAY BREAK!
4. Replace the Sample Detector Circuit board (335901-6034 [UV] or 335901-6044 [Vis]).
NOTE: The detectors are specific to the instrument type (UV vs. Vis) so you cannot interchange
the two UV Detector boards and the Vis Detector boards by simply changing the jumper.
Ensure that you have the correct board and the correct jumper configuration:
W1 Jumper Settings:
Vis Sample Detector = pins 2 and 3 are jumpered
UV Sample Detector = pins 1 and 2 are jumpered and pins 3 and 4 are jumpered
UV Reference Detector = pins 1 and 2 are jumpered and pins 3 and 4 are jumpered
W2 Jumper Settings:
Sample Detector (Vis and UV) = W2 pins are jumpered together
5. Insert the ribbon cable into its connector making sure the No. 1 wire on the cable is at the No. 1
position on the connector and lock the connector.

NOTE: Orient the ribbon cable so that the striped (non-conductive) side of the cable connector is
facing the Circuit board.
NOTE: Ensure that the end of the cable is inserted all the way into the connector. No part of the
exposed leads should be visible when the cable is locked into the connector.
6. Position the Circuit board in its mounting position making sure the holes on the Circuit board fit
over the two dowel pins.
9. Position the cover over the Sample Detector Circuit board and lens.
10. Insert and tighten the lower screw and the one upper screw, which is not common to the Beam
Splitter and turret cover.
11. After all replacements are complete for mirrors, light source, grating, and lenses, follow the optical
alignment procedures in “Optical Alignment."
12. When all other work is complete, replace the Beam Splitter and turret cover (refer to “Beam splitter
and turret cover ").
Removal
1. Remove the Beam Splitter and turret cover (refer to “Beam Splitter and turret cover").
3. Remove the two screws securing the top of the Reference Detector cover.

4. Remove the remaining lower screw securing the Reference Detector cover.
5. Lift the Reference Detector cover off the base and set aside.
7. Remove the two screws securing the Reference Detector Circuit board to the base.
8. Carefully pull the Reference Detector Circuit board away from its locating dowel pins and remove it
from its mounting position.
9. Remove the ribbon cable from its connector on the Reference Detector Circuit board.
CAUTION: NEVER TOUCH THE LENS WITH YOUR FINGERS. ALWAYS USE AN
UNCOATED LENS TISSUE, OR SOFT COTTON CLOTH, OR CLEAN LAB

NOTE: The Opto switch, turret motor, and turret Shaft may each be removed independently
without removing the other two items, if so desired.
Removal

1. Remove the Beam Splitter and turret cover (refer to “Beam splitter and turret cover ").
3. Turn the unit over and unplug the connector for the Opto switch /turret motor from the Main Circuit
board.
4. From the top side, remove the two screws, which secure the turret Opto Switch to the base.
5. Fit the wire connector up through the hole in the base being careful not to damage any of the other
wires passing through the hole and set the switch aside.
7. From the top, remove the two screws securing the turret motor to the base.
9. Fit the wire connector up through the hole in the base being careful not to damage any of the other
wires passing through the hole if not done in the previous step and set the motor aside.
10. Remove the Main Circuit board (refer to “Main board Connections").
11. Remove the turret shaft by looseing the set screw that holds the gear in place and removing the
gear. Lift the shaft out from the top of the instrument.
12. From the top, remove the E-clip securing the turret shaft in its bushing.
13. From the bottom, grasp the driven gear and carefully pull the shaft out through the bushing.
NOTE: On single cell units, the shaft is cemented in place and used as a guide only.

2. Pass the turret shaft with attached driven gear up through the bushing from the bottom of the unit
until the groove for the E-clip is accessible above the top of the bushing. If the turret motor was not
removed. It may be necessary to rotate the gear slightly to make sure the teeth of the driven gear
mesh with the teeth of the drive gear on the motor shaft.
5. Pass the turret motor shaft through the hole in the base and push it all the way down till the motor
is flush against the base. DO NOT FORCE THE MOTOR! It may be necessary to rotate the motor
slightly to make sure the teeth of the drive gear on the end of the motor shaft mesh properly with
the teeth of the driven gear.
6. Insert and tighten the two screws, which secure the motor to the base.
7. Pass the connector end of the motor wires through the hole in the base making sure the other
wires passing through the hole are not damaged.
8. If all other work is complete, replace the Main Circuit board (refer to "Main board Connections").
9. Attach the connector on the end of the motor wires to the Main Circuit board.
11. Insert and tighten the two screws, which secure the turret Opto Switch to the base.
12. Attach the connector on the end of the switch wires to the Main Circuit board.
Light Source (335923-000 [Vis Only], 335902-146S [UV-Vis Only] and 335908
[Spectronic GENESYS 6 Only])
NOTE: There are two possible light sources for the Spectronic GENESYS 10
Spectrophotometer: xenon (UV-Visible) or tungsten (Visible only). Each type is unique to
a specific instrument they cannot be interchanged. The Vis lamp is supplied as an
assembly already aligned on its mounting brackets and is replaced as a complete
assembly. The UV Lamp is supplied alone and requires optical alignment after replacing.

Removal
NOTE: The xenon light source bracket has two screws and two dowel pins.
2. Remove the screws, which secure the light source bracket to the base.
3. Lift the light source bracket off the dowels and set aside.
Replacement
1. Remove the xenon lamp from the bracket. Mount the new xenon lamp on the bracket.
3. Insert and tighten the screws holding the light source bracket to the base.
4. Turn the unit over and attach the light source wire connector to the Main Circuit board.
Mirrors
Removal
1. Remove the Beam Splitter and turret cover (refer to “Beam splitter and turret cover ").

NOTE: Each mirror is secured to the base using the two outer screws. The center screw is used
for vertical alignment of the mirror.
3. Remove the two screws securing the mirror to the base and set the mirror aside.
Replacement
1. Insert the two screws, which secure the mirror to the base. They do not have to be tightened at this
step.
2. After all replacements are complete for mirrors, light source, grating, and lenses, follow the optical
alignment procedures in "Optical Alignment."
3. When all other work is complete, replace the Monochromator cover (refer to “Monochromator ").
4. When all other work is complete, replace the Beam Splitter and turret cover (refer to “Beam splitter
and turret cover ").
Removal
1. Remove the Beam Splitter and turret cover (refer to “Beam splitter and turret cover ").
3. Loosen the locking screw on the clamp collar around the grating assembly.

4. Carefully lift up on the grating assembly to pull it off the grating motor shaft. The plastic neck of the
grating assembly may come out of the clamp collar as it is being pulled upward.
5. If it is not already off, remove the clamp collar from the grating assembly and set it aside for later
use on the new grating assembly.
Replacement
1. Position the clamp collar around the plastic neck of the grating assembly with the clamp screw
loose.
2. Slide the grating assembly and collar onto the grating motor shaft making sure the collar stays in
place around the neck of the grating assembly.
3. Slip the spacing tool (from the alignment kit) between the grating and the grating motor to provide
0.5mm space. This space prevents any friction between the grating mount and the top of the motor.
5. After all replacements are complete for mirrors, light source, grating, and lenses, follow the optical
alignment procedures in Optical Alignment.
6. When all other work is complete, replace the Monochromator cover (refer to “Monochromator ").
7. When all other work is complete, replace the Beam Splitter and turret cover (refer to “Beam splitter
and turret cover ").

2. Turn the instrument on its side as shown and disconnect the wire connector for the grating motor
from the Main Circuit board.
3. From the topside of the instrument, remove the two screws securing the grating motor to the base.
4. Carefully remove the grating motor so the shaft comes out of the base and set the motor aside.
Replacement
1. From the bottom of the unit, insert the shaft of the grating motor through the hole in the base and
turn the grating motor so it’s threaded mounting holes are aligned with the mounting holes in the
base.
2. Insert and tighten the two screws used to secure the grating motor to the base.
3. Attach the wire connector for the grating motor wires to the Main Circuit board.
4. Replace the grating assembly on the grating motor shaft (refer to “Grating assembly").

from the Main Circuit board.

3. From the bottom of the instrument, remove the three screws securing the Power Supply bracket to
the base. Leave the Power Supply bracket in place; it only needs to be loose enough to allow room
to remove the grating motor Block assembly.
4. Remove the two 8-32 screws that secure the grating motor block assembly to the base.
5. Carefully remove the grating motor Block assembly so the shaft comes out of the base and set the
motor aside.
Replacement
1. From the bottom of the unit, insert the shaft of the grating motor Block assembly through the hole in
the base and turn the grating motor Block assembly so it’s mounting holes are aligned with the
mounting holes in the base.
2. Insert and tighten the two 8-32 screws used to secure the grating motor Block assembly to the
base.
3. Attach the wire connector for the grating motor wires to the Main Circuit board.

4. Replace the grating assembly on the grating motor shaft (refer to “Grating assembly").
Removal
2. Unplug the power and ribbon cable from the back of the drive, note orientation.
3. From the bottom of the instrument, remove the four screws securing the floppy drive bracket to the
base.
4. Remove the two flathead screws that secure the floppy drive to the bracket
Replacement
1. Insert and tighten the two flathead screws used to secure the floppy drive to the bracket.
5. Replace the floppy drive and bracket on base and secure it with the four screws that were removed
earlier.
2. Plug the power and ribbon cable back into the connectors at the back of the drive, note orientation.

Removal
1. Disconnect the wire connector for the Power Supply wires from the top of the Main Circuit board.
2. Turn the instrument over and remove the screw securing the ground wire. This screw is one of the
six screws used to secure the Main Circuit board. Note how the wire is positioned through the hole
in the sidewall of the Power Supply cavity.
3. Remove the three screws securing the Power Supply assembly to the base.
Replacement
1. Insert the Power Supply into the base. Make sure the ground wire is properly routed through the
hole in the sidewall of the base cavity.
2. Insert the three screws used to secure the Power Supply to the base.

3. Position the ground wire over its mounting hole and insert and tighten the mounting screw.
4. Attach the wire connector to the top of the Main Circuit board.
Main (CPU) Circuit board (335901-6024S [UV-Vis Only]; 335901-6064S [Vis Only])
Removal
1. Disconnect all wire connectors on the top and end of the Main Circuit board, accessible at the rear
of the instrument through holes in the base casting.
2. Turn the instrument over and remove the seven screws attaching the board to the base.
3. Note how the ribbon cables are routed around the instrument standoffs.
4. Carefully lift the Circuit board away from the base and disconnect the balance of the wire
connectors.
Replacement

1. Attach each of the bottom and edge cable connectors to their appropriate position on the Main
Circuit board.
2. Carefully position the board in its mounting location. Make sure the mounting holes in the board
line up with the mounting holes in the base. Carefully route the ribbon cables as necessary to make
sure they will not be damaged when the covers are replaced, or pinched under the board.
4. Turn the unit over and attach the remainder of the wire connectors to the top and end of the Main
Circuit board.
5. If the firmware revision of the new board is ≤ 1.030, then replace the ROM and flash it to the
current version. If the firmware revision of the new board is > 1.030, then the firmware must flashed
into the ROM (see the instructions for GenFlash). Recalibrate the instrument.
Display
Removal
3. Remove the four screws holding the display to the top cover and set aside.

Replacement
1. Place the new display in the top cover. Make sure the cables are at the bottom of the display as it
is located in the cover and the connector is towards the front of the instrument.
Keypad
Removal
2. Disconnect the ribbon cable from the connector on the Main Circuit board.
3. Carefully peel the keypad off the top cover and pull the ribbon cable through the hole in the cover.
Clean off any excess adhesive.
Replacement
1. Start the ribbon cable through the hole in the top cover.
CAUTION: AFTER THE BACKING IS REMOVED, DO NOT LET THE KEYPAD TOUCH
THE TOP COVER OR ANYTHING ELSE UNTIL IT IS READY TO BE
PRESSED IN POSITION OR THE KEYPAD MAY BE DAMAGED TO WHERE
IT CANNOT BE USED.
2. Peel the backing off the keypad, carefully position it over the top cover, and press it into position.

WARNING
Turn off the instrument and disconnect the power cord from the outlet before installing the
Internal Printer.
1. Loosen the captive screw (#1, Figure 5.25 Printer Installation) on the lamp door by rotating it
counterclockwise about ¼ turn.
3. Use a pen or screwdriver to lift the tabs holding the door to the hinge (#2 and #3, Figure 5.25
Printer Installation).
5. Remove the printer (already installed on the printer door) from its packing.
8. Connect the wires (#4, Figure 5.25 Printer Installation) and press them into place with a small
screwdriver. There is only one way that the connector will fit. Look closely at the end of the
connector, it has a slight D shape, or make sure that the side of the connector with the shiny metal
contacts showing, faces away from the printer and towards the plastic door. In the figure above, the
side of the connector with the shiny metal contacts showing faces down.
10. Install the printer door by sliding it back onto the hinge (#3, Figure 5.25 Printer Installation).
12. Tighten the captive screw (#1, Figure 5.25 Printer Installation) on the printer door to hold it securely
in place.
13. Load the paper into the printer (see instructions in Setting up the Instrument in the operator
manual).

Chapter 6 Optical Alignment
WARNING
When servicing your Series spectrophotometer, always observe proper safety precautions

DO NOT USE STEEL WOOL, EMERY CLOTH, SAND PAPER, OR FILES WHEN CLEANING
THE INSTRUMENT.
To perform the optical alignment, start with the power off. Remove the main cover, the mono cover, the
spill tray, and the filter wheel. Then turn on the power to the unit. The lamp on the Visible only models will
remain lit.

Optical Alignment
Instruments boot differently depending on instrument model, version, and if it is calibrated. All that is
required to align the optics is to light the lamp. The lamp is always lit in Visible units.
2. Press ENTER to flash the lamp for 15-second intervals after power on.

The four mirrors used in the Spectronic GENESYS 10 spectrophotometer are each unique and must only
be replaced by the same mirror at a specific location. However, all four mirrors mount and adjust in the
same manner. To tell the difference:
There are two screws (one at each end of the foot), which mount the mirror to the base (refer to
Figure 6.1). There is enough clearance in the mounting hole to provide some rotational movement of the
mirror. This movement is used to adjust the horizontal (rotational) alignment of the mirror.
A third screw is located behind the mirror. It holds down the spring-loaded mounting tab. This screw is
used to adjust the vertical tilt of the mirror.

Optical Alignment
1. Make sure the proper mirror has been selected for the location and is oriented with the mirrored
surface facing in the correct direction as shown in Figure 6.2. (All mirrors face in toward the center
of the instrument.)
2. Insert the two mounting screws through the foot of the mirror into the base. Snug the mirrors
slightly but do not tighten. The mirror should be able to be rotated in the hole with a slight
resistance. The screws must be loose enough so that the mirror will slide under the screw. Never
force or twist the mount to align it.
5. Tighten or loosen the tilt adjustment screw as necessary until the vertical alignment of the mirror is
proper.
Alignment Sequence
The various components of the optical path and their locations are shown in Figure 6.2. In addition to any
replacement components, which may be required, the following procedure requires the use of the
Spectronic GENESYS 10 Service Alignment Tool Kit [335901-400], which includes the following
components:
• 335901-401 Mirror B2 Target
• 335901-408 Dental Mirror
• 335901-403 zero order grating Tool
• 335901-406 Spectronic GENESYS 10 Sample Target
• 335901-407 Spectronic GENESYS 10 Mirror Adjust Tool

WARNING
When in operation, certain internal components, such as the lamp and Power Supply,
become hot. Avoid touching these areas. Failure to observe this warning may result in burns.
WARNING
High voltage is present in the instrument when power is turned on. Be very careful to avoid
any voltage carrying contacts while performing these alignment procedures. Failure to heed
this warning may result in serious personal injury including the potential for severe burns,
heart failure, or death.
WARNING
Never stare directly at the light source or the reflected beam from the light source. Always
wear UV protection in the form of safety glasses or goggles. Failure to heed this warning
could result in potential eye damage.
Remove the Monochromator cover, the Beam Splitter and turret cover, and the filter wheel (refer to
“Disassembly and Replacement").

Optical Alignment
1. Hold the dental mirror from the Alignment Tool Kit in the area normally occupied by the filter wheel
behind the entrance slit.
2. Align the source mirror A horizontally and vertically until the beam is centered on the entrance slit
as observed using the dental mirror (refer to “Mirror Adjustments").
1. Loosen and remove the two screws securing the limit switch to the base. Move the limit switch to
one side so it will not interfere with the installation of the zero order grating Holder tool.
2. Lock the tool in the ‘open’ position by compressing the spring until the pin can be inserted into the
locking hole in the tool.
3. Position the zero order grating Holder tool so that the pin in the base of the tool can be inserted into
the rear screw hole where the limit switch was installed and the hole in the tool base is located over
the front screw hole. Position the grating arm in the L-shaped arm of the tool. Push the tool to the
right as shown in Figure 6.3 when tightening the screw.
4. Place the Mirror B2 Target over the reflective surface of the B2 mirror.
5. Align the B1 Monochromator mirror horizontally and vertically until the beam is centered on the
Mirror B2 Target (refer to “Mirror Adjustments").

NOTE: Check the alignment by loosening the arm in the zero order grating Holder tool
(compress the spring on the zero order grating Holder until the pin can be inserted into
the locking hole, and the tool is locked in the ‘open’ position) and moving the grating arm
to the right. A spectrum will be reflected from the grating onto the entrance slit. Ensure
that this spectrum is centered vertically on the slit. If this requires adjustment of the
mirror, then retighten the zero order grating Holder tool, readjust vertically, and ensure
that the light beam is still centered on the target.
2. Align the B2 Monochromator mirror horizontally and vertically until the beam is centered on the Exit
Slit (refer to "Mirror Adjustments") and on the target on Mirror C then remove the target from
Mirror C.
1. Orient the Sample Target in position on the Cell Holder Platform so that the rectangular target is
facing the light beam. Secure the Sample Target with its captive screw. Install the Platform with the
Sample Target into the sample compartment.
2. Align the Sample Mirror C horizontally and vertically until the beam is centered on the Sample
Target (refer to "Mirror Adjustments").
3. When the alignment has been completed, remove the Cell Holder Platform. Remove the Sample
Target from the Cell Holder Platform and reinstall the previously installed cell holder onto the
Platform.
4. Compress the spring on the zero order grating holder until the pin can be inserted into the locking
hole, and the tool is locked in the ‘open’ position.
5. Loosen and remove the screw securing the zero order grating Holder tool.
7. Reinstall the Limit Switch, the filter wheel, the Monochromator cover, the Beam Splitter and turret
cover.
Note: After any optical change, alignment, or optical component replacement, the instrument
must be calibrated with the proper standards using the Service Diagnostic. Instructions
are included with the software.

Alignment Procedure:
4. Gently tap the cell on a padded surface to ensure that there are no bubbles in the light path.
1. Use Service Diagnostics to continuously pulse the xenon lamp and to access zero order.
2. Place a piece of clean white paper (like the back of a calling card) up against the sample detector
window so that the clean white side faces the single cell holder.
4. Observe the intensity of light reflecting off the white paper as adjustments of Mirror C are made. Set
for maximum uniform light image intensity on the white paper.
Final Check:
5. Gently tap the cell on a padded surface to ensure that there are no bubbles in the light path.
6. Insert the cell in the cell holder and zero the instrument.
7. Remove and re-insert the microcell in exactly the same position several times.
An additional check is to turn the microcell 180 degrees (reverse direction), reinsert the cell and compare
the forward/reverse direction agreeability. There is no specification on this; however, it should have
similar repeatability.
Perform the above ‘Final Check’ at several wavelengths, especially at 260nm and 280nm.
Note: The ‘axial’ position of the square cuvette holder also affects the reading/repeatability. It
may be necessary to loosen the screw that secures the cuvette and turn it to ensure that
the light beam passes straight through the cell windows on BOTH sides of the cuvette
without hitting the inner side wall of the cell’s sample chamber.

Chapter 9 SERVICE DIAGNOSTICS SOFTWARE
The Service Diagnostic software can be used to both calibrate and troubleshoot the Spectronic
GENESYS 10 Series of spectrophotometers, including the Spectronic GENESYS 10 Vis, Spectronic
GENESYS 10 UV, Spectronic GENESYS 10 UVscanning, Spectronic GENESYS 6, Spectronic BioMate 3
and Spectronic GENESYS 10 Bio models. Calibration requires a didymium filter. The program can be
used to exercise motors independently, check gain settings on the detector(s), as well as calibrate the
instrument. The Spectronic GENESYS 6 model auto-calibrates at power up, so there is no option to
calibrate it here.
The instrument can be reset to default by holding down the ↓ and ENTER keys at the same time on the
instrument key pad while turning on power and then selecting the desired menu option.

Alignment Tool
1. Used to check if limit switch is inoperative and to check/set the zero order. Should be performed
before physically aligning any of the Mirrors.
2. If the limit switch is inoperative, the grating arm will just bounce back and forth since it cannot ‘find’
the limit switch.
3. If the system is operating correctly, the white beam will be positioned to the left of the exit slit.

1. Used to check alignment on a calibrated instrument. The control will be enabled after seek Limit
Switch is performed. It moves the grating a set number of steps from the limit switch to where the
instrument thinks zero order is located. The Adjust Zero Order slider allows a +/- 100-step
adjustment.
3. If the zero order is correctly set, the light beam will be centered on the exit slit.
4. If not, left-click and drag the Adjust zero order Slider to adjust the position of the light beam
between flashes (should see maximum energy value on window). The controls are disabled while
the instrument is flashing. Un-check the Flash box or grab the slider when the controls are enabled
to change the grating position. Do not turn the instrument off while flashing. Close the window and
then turn off the instrument.
5. If the beam is too far from the slit to allow this adjustment, then perform the physical mirror
adjustment with the ZOGH tool.
Flash Xenon
1. This control is used to flash the Xenon lamp for optical alignment.
2. Click Flash Xenon to start flashing the lamp at the maximum allowed rate.
3. Click OK to stop.
1. The Sample Energy and Reference Energy readouts display the raw counts from the A/D converter
minus dark current for the respective channel. The capacitor and voltage settings are used to
compensate for different energy levels at different wavelengths, or different amounts of light hitting
the detector(s). They are normally set automatically when the instrument is zeroed, or measure
blank is pressed. The detector circuit uses an integrator circuit. Changing the number for the

Capacitor or Voltage simply selects a different capacitor or resistor value to use in the feedback
loop.
2. Use the Detector board Diagnostic screen to test for proper detector operation. The higher the gain
number, the higher the Energy level should be. The reading will saturate around 6,300,000 counts.
For each capacitor or resistor change there should be a corresponding change in the Sample
Energy or Reference Energy level. If there isn’t a corresponding change, and the reading isn’t
saturated, there is a problem with the Main board, detector, or ribbon cable. A simple way to test
the gain settings is to set the instrument wavelength to a low energy point, like 850nm and try each
gain setting.
3. Can be used when the covers have been removed, but the sample compartment must be covered
with a light-tight covering (black cloth).
5. For the UV model, the Sample Energy and the Reference Energy will be roughly equal, after the
instrument is zeroed (see examples on next page).

Chapter 10 RS232 Commands
The RS232 Commands allow a computer to control the instrument for data acquisition, or
troubleshooting. Use any standard terminal emulation program to issue a command. Settings are:

Whenever a Display (either the older-style Hantronix with black wires and the
13-pin connector, or the newer-style EDT with white wires and the14-pin
connector) needs to be replaced, order the 335901-657S Display Kit.
Please note that the 335901-657S Display requires software revision (1.103
for the Spectronic GENESYS 10, and 1.001 for the Spectronic BioMate 3). If

PROBLEM: 1. Earlier revision of firmware will not work with new EDT display (335901-
657S).
2. Error in Performance Validation test – Internal Wavelength tolerance (is
±1nm at 883nm, should be ±2nm).
SYMPTOM/ Problem 1 - can cause the display to be blank when the unit is turned on (usually
CAUSE: after powering-down an instrument that has been on beyond the warm-up period).
Problem 2 - can cause the instrument to fail while still within the published
specifications.
SOLUTION: Release of new firmware: 1.103 for Spectronic GENESYS 10; 1.001 for
Spectronic BioMate 3
The new firmware will work with the new EDT display (see TSB S-391), and has the
correct Internal Wavelength Tolerance of ±2nm at 883nm.
If the unit has the new EDT display which shows the problem above, the unit should
be sent to the Service depot for upgrading since the upgrade will require the
installation of the ROM IC and wavelength recalibration using the appropriate
standards and software.

SYMPTOM/ The ‘flashable’ ROM was not implemented in the production of the Spectronic
CAUSE: GENESYS 10 until December 2000. The firmware in the units made before this
date can only be upgraded by the physical replacement of the ROM.
SOLUTION: ROM Upgrade Kits have been developed to upgrade the pre-December 2000 units
to ‘flashable’ ROMs
There are two part numbers for the Spectronic GENESYS 10 ROM replacement:

Caution Do not begin this procedure unless you have a Spectronic GENESYS 10
Service manual (Catalog number 335901-10020) and a certified wavelength
filter.
This upgrade procedure requires the use of a didymium filter (certified or
non-certified) for the wavelength calibration of the instrument and the use
of a traceable wavelength standard for final testing and certification after
calibration. (This may be the same didymium filter used for calibration if a
certified didymium filter was used for the calibration.)
• Thermo Electron Corporation sells wavelength filters:
• Filter Set (catalog number 942318503111) which includes a traceable holmium filter (can be used
only for final testing & certification) and a traceable didymium filter (can be used for both
calibration and certification)
• SPECTRONIC Standards (catalog number 333150) which includes a traceable wavelength filter
(can be used only for final testing & certification)
• Non-certified didymium filter (catalog number 333159) for only the wavelength calibration
• Performing this upgrade will cause any stored tests to be deleted due to improvements in the
software and resultant data structures. Therefore, we recommend printing or otherwise recording your
test setup screens for greater ease in re-programming them.
• Observe proper ESD procedures.
• Use appropriate tools (e.g., PLCC extractor tool).
• A Personal Computer is required to perform this procedure, along with a working RS232 serial port
and interface cable (Part #335942 or equivalent*).
• The Spectronic GENESYS 10 Service Diagnostic software Rev. 1.40 or higher (included in your
service manual, or sold separately as Part # 335901-10005) is also required to perform this upgrade.
Procedure
5. Remove the cover gently, so as not to stress the ground wire connection.
6. Using a PLCC ROM extractor tool, remove the ROM (marked with a label). The board is marked
U39 next to the socket. This is the only Integrated Circuit (IC) on the Main Circuit board that is
socketed and can be removed. Do not attempt to remove any other components, including
connectors or jumpers. (See Figure 11.1 Close-up photo of the socket and ROM [on next page].)

7. When installing the new ROM, take care to note the orientation of the notch and the “dot” on the
chip, and line them up with the notch in the socket and the dot printed on the Circuit board (see
Figure 11.1). Note that the label may be in any orientation- do not use the label to determine the
orientation of the IC.
8. After ensuring proper alignment, press the ROM gently into the socket and make certain that it is
fully seated.
Caution: Improper insertion will damage the new ROM and possibly the instrument!
9. After installation is complete, replace and secure the bottom cover and turn the unit right-side-up.
10. Connect Power and turn on the instrument.
11. When prompted, press F1 or F2 (first or second unmarked key from the left [under the display])
for a UV or Visible-only model, respectively.
12. After the unit boots up, connect the instrument to a PC running the Spectronic GENESYS 10
Service Diagnostic application package (19200,N,8,1), and follow the calibration instructions in
the Service manual.
Note: You will need to re-enter the serial number into the instrument using the Service
Diagnostic application.
